چکیده

Background “Inside-Outside” hypothesis Cells located on the inside of the 16-cell morula differentiate into the inner cell mass (ICM) of the blastocyst while cells on the outside of the morula differentiate into the trophectoderm (TE) If a cell were moved to a different location in the morula, it would differentiate into a different kind of cell Hypothesis and corollary assumed that, for a time, all 16 cells in morula were identical. Later proved to be false; as soon 16-cell morula exists, the cells within the morula can be divided into 2 groups Apolar cells – smaller, label on surface uniformly, sparse but uniform distribution of microvilli, more adhesive, located on the inside Polar cells – larger, polar distribution of microvilli, less adhesive, tend to envelope aggregated apolar cells, located on the outside Recently outer cells were injected with horseradish peroxidase (HRP) and found that in 44% of embryos analyzed they contributed to both ICM and TE, and 53% contributed only to the TE

Simultaneous regulation of miR-451 and miR-191 led to erythroid fate decision of mouse embryonic stem cell

Objective(s): Various microRNAs (miRNAs) are expressed during development of mammalian cells, when they aid in modulating gene expression by mediating mRNA transcript cleavage and/or regulation of translation rate. miR-191 and miR-451 have been shown to be critical regulators of hematopoiesis and have important roles in the induction of erythroid fate decision. So, the aim of this study is inve...

Anti-Oxidative and Anti-Apoptotic Effects of Apigenin on Number of Viable and Apoptotic Blastomeres, Zona Pellucida Thickness and Hatching Rate of Mouse Embryos

Objective Apigenin is a plant-derived compound belong to the flavonoids category and has the protective effects on different cells. The aim was to evaluate the effect of apigenin on the number of viable and apoptotic blastomeres, the zona pellucida (ZP) thickness and the hatching rate of preimplantation mouse embryos exposed to H2O2 and actinomycin D. MaterialsAndMethods 420 two-cell embryos we...

Mouse blastomeres acquire ability to divide asymmetrically before compaction

The mouse preimplantation embryo generates the precursors of trophectoderm (TE) and inner cell mass (ICM) during the 8- to 16-cell stage transition, when the apico-basal polarized blastomeres undergo divisions that give rise to cells with different fate. Asymmetric segregation of polar domain at 8-16 cell division generate two cell types, polar cells which adopt an outer position and develop in...

The Influence of Light on Apoptosis in Preimplantation Mouse embryos in vitro

Background: In vitro culture of mammalian embryos can slow or stop growth completely. This may be due to the medium used, pH, temperature, or light. There is considerable concern about the harmful effect of light in the laboratory environment. Cell number and apoptosis are useful parameters that indicate embryonic development and health. In this study, we assessed these two factors in the blas...

Inactivation of aPKCλ Reveals a Context Dependent Allocation of Cell Lineages in Preimplantation Mouse Embryos

BACKGROUND During mammalian preimplantation development, lineage divergence seems to be controlled by the interplay between asymmetric cell division (once cells are polarized) and positional information. In the mouse embryo, two distinct cell populations are first observed at the 16-cell stage and can be distinguished by both their position (outside or inside) and their phenotype (polarized or ...
